2000 Liberty 2.5ltr has a misfire and loss of power under load wife fuelled car with E10 91 octane fuel emptied tank after 400k ran a tank of 95 octane and injector cleaner, seemed to correct problem has now developed a misfire and loss of power under acceleration not sure what to look for thinking either fuel blockage or spark plugs or even coil plugs It hesitates has a miss then runs fine

Any suggestions will be appreciated

Try resetting the ecu by unplugging the battery, then getting rid of the charge by pressing the brake pedal a few times. Then reattach the battery to see if it makes a difference....

Pull a spark plug and check its condition.
For the price of a set of new plugs, you'd be best just doing a plug change and crossing them off the list of suspects.
